Nicotine-derived JV-Nitrosamines and Tobacco-related Cancer: Current Status and Future Directions1

چکیده
Although per capita use of cigarettes among adults (18 years of age and over) has declined from 4345 in 1963 to 3494 in 1983, there are still about 53 million cigarette smokers in the United States (89, 92). Six hundred thirty billion cigarettes were con sumed in 1983. An alarming development is the increasing prevalence of snuff dipping. Snuff dipping is the practice of extracting juices from a pinch of moist fine-cut chewing tobacco, placed between the cheek and the gum. The general popularity of snuff dipping, especially among young people, is a relatively recent trend but women in the southeastern parts of the United States have been snuff dippers for many decades. A recent case-control study showed that the relative risk of oral cavity cancer among white nonsmoking, snuff-dipping women was 4.2. Among chronic users, the relative risk of cancers of the gum and oral cavity approached 50 (96). In 1983, consumption totaled 21,000 tons of snuff tobacco and 39,000 tons of chewing tobacco (89). Presently, there are an estimated 7 million snuff dippers in the United States (80). The main reason for the continued use of tobacco, in spite of its well-known adverse health effects, is dependence on nicotine (64). This compound is the major alkaloid2 in United States

Tobacco-specific nitrosamines, an important group of carcinogens in tobacco and tobacco smoke.
Tobacco-specific nitrosamines are a group of carcinogens that are present in tobacco and tobacco smoke. They are formed from nicotine and related tobacco alkaloids. Two of the nicotine-derived nitrosamines, NNK and NNN, are strong carcinogens in laboratory animals. They can induce tumors both locally and systemically. The induction of oral cavity tumors by a mixture of NNK and NNN, and the orga...
متن کاملNicotine: Carcinogenicity and Effects on Response to Cancer Treatment – A Review
Tobacco use is considered the single most important man-made cause of cancer that can be avoided. The evidence that nicotine is involved in cancer development is reviewed and discussed in this paper. Both tobacco smoke and tobacco products for oral use contain a number of carcinogenic substances, such as polycyclic hydrocarbons and tobacco-specific N-nitrosamines (TSNA), which undoubtedly contr...
متن کاملTransgenic and Mutation-Based Suppression of a Berberine Bridge Enzyme-Like (BBL) Gene Family Reduces Alkaloid Content in Field-Grown Tobacco
Motivation exists to develop tobacco cultivars with reduced nicotine content for the purpose of facilitating compliance with expected tobacco product regulations that could mandate the lowering of nicotine levels per se, or the reduction of carcinogenic alkaloid-derived tobacco specific nitrosamines (TSNAs). A berberine bridge enzyme-like (BBL) gene family was recently characterized for N. taba...
متن کاملExposure to nicotine and carcinogens among Southwestern Alaskan Native cigarette smokers and smokeless tobacco users.
BACKGROUND The prevalence of tobacco use, both cigarette smoking and smokeless, including iqmik (homemade smokeless tobacco prepared with dried tobacco leaves mixed with alkaline ash), and of tobacco-related cancer is high in Alaskan Native people (AN). To investigate possible mechanisms of increased cancer risk we studied levels of nicotine and tobacco-specific nitrosamines (TSNA) in tobacco p...
متن کاملAgonist and antagonist effects of tobacco-related nitrosamines on human α4β2 nicotinic acetylcholine receptors
Regulation of the "neuronal" nicotinic acetylcholine receptors (nAChRs) is implicated in both tobacco addiction and smoking-dependent tumor promotion. Some of these effects are caused by the tobacco-derived N-nitrosamines, which are carcinogenic compounds that avidly bind to nAChRs. However, the functional effects of these drugs on specific nAChR subtypes are largely unknown. By using patch-cla...
